Accurate dosage calculation is fundamental in peptide research. This guide details the conversion formulas between milligrams, international units, and insulin syringe graduations, with concrete examples for the most common research peptides.

Fundamental principles

Before any calculation, it is necessary to understand the three variables involved:

  • Quantity of peptide: mass in mg contained in the vial (e.g., 5 mg, 10 mg)
  • Reconstitution volume: quantity of bacteriostatic water added (e.g., 1 mL, 2 mL)
  • Resulting concentration: mass/volume ratio (e.g.: 5 mg/mL, 2.5 mg/mL)

Understanding the insulin syringe

U-100 insulin syringes are the standard tool in peptide research due to their low-volume accuracy:

Important

The "units" (IU) on an insulin syringe are NOT international peptide units. They are simply volumetric graduations: 1 IU = 0.01 mL = 10 microlitres.

Universal calculation formula

The basic formula for all peptide assay calculations:

Volume to be taken (mL) = Desired dose (mg) / Concentration (mg/mL)

And to convert to syringe graduations:

Ticks to be taken = Volume (mL) x 100

Examples of common calculations

Example 1: BPC-157 (5 mg vial)

Reconstruction 5 mg in 2 mL BAC Concentration = 2.5 mg/mL (= 2500 mcg/mL)
Target dose: 250 mcg 0.25 mg / 2.5 mg/mL = 0.1 mL = 10 IU (ticks)
Target dose: 500 mcg 0.5 mg / 2.5 mg/mL = 0.2 mL = 20 IU (ticks)

Example 2: Ipamorelin (5 mg bottle)

Target dose: 200 mcg 0.2 mg / 2 mg/mL = 0.1 mL = 10 IU (ticks)

Example 3: Semaglutide (3 mg vial)

Common laboratory errors

  • Confusion between insulin IU and peptide IUThe syringe graduations are VOLUME measurements, not biological potency measurements.
  • Forget the dead volumeInsulin syringes have a minimal dead volume (~5 mcL), but there is also a dead volume for removable needles.
  • Reconstruction too hastyAlways let the water run down the side of the bottle; never inject directly onto the lyophilized powder.
  • Air bubblesAn air bubble in the syringe reduces the actual volume drawn — always prime before measuring
  • Misreading the graduations: read at the level of the lower meniscus, syringe at eye level

Good laboratory practices

  • Always note the concentration obtained on the bottle after reconstitution.
  • Use the smallest syringe suitable for the volume to be drawn (0.3 mL if possible).
  • Store the reconstituted vials in a refrigerator (2-8°C).
  • Do not reuse insulin syringes (single use only)
  • Disinfect the bottle cap with alcohol before each use.
  • Document each sample (date, volume, concentration)

To simplify calculations, reconstitute to obtain "round" concentrations. For example: 5 mg in 2 mL = 2500 mcg/mL, which gives exactly 250 mcg per 10 ticks. Simple and error-free.
