Le WORDS-CMOTS-C (Mitochondrial Open Reading Frame of the twelve S rRNA type-C) is a 16-amino-acid mitochondrial derivative peptide (MDP) encoded in the 12S rRNA gene of the mitochondrial genome. Discovered in 2015 by Dr. Pinchas Cohen's team at the University of Southern California, it represents a major advance in understanding mitochondria-nucleus communication and the regulation of energy metabolism. MOTS-C is one of the few known peptides encoded in the mitochondrial genome and to exert systemic biological effects.
Molecular mechanism of action
MOTS-C exerts its effects via several metabolic signaling pathways: (1)AMPK activation(AMP-activated protein kinase) — central regulator of cellular energy metabolism, involved in mitochondrial biogenesis and fatty acid oxidation; (2)Nuclear translocation— during metabolic stress, MOTS-C migrates to the nucleus where it directly regulates the expression of genes related to metabolism; (3)Regulation of the folate cycle - AICAR— inhibits the AICAR pathway to rebalance cellular metabolic status; (4)Improved insulin sensitivity— via the activation of the GLUT4 transporter and the suppression of insulin resistance induced by saturated fatty acids.
