Le PT-141 10mg(Bremelanotide) is a synthetic cyclic heptapeptide derived from Melanotan 2, of which it is the deamidated metabolite. Unlike MT2, it does not induce significant pigmentation: its affinity is concentrated on melanocortin receptors.MC4R et MC3Rexpressed in the hypothalamus and central nervous system. It is the first melanocortinergic agonist to have been the subject of clinical trials on sexual function, making it a reference tool for research on the central pathways of desire and arousal.
Molecular mechanism of action
The PT-141 operates on several levels: (1)MC4R agonism— activation of hypothalamic neurons involved in the sexual response, via a central pathway independent of the vascular system (unlike PDE5 inhibitors); (2)MC3R agonism— modulation of energy homeostasis and anti-inflammatory pathways; (3)dopamine releasein the medial preoptic area, documented in animal models; (4)low activity MC1R— limited pigmentation compared to MT2; (5)cyclic structure— superior stability and half-life compared to endogenous α-MSH.
